Abstract
The side effects of anticoagulant and thrombolytic treatment are the reason of alternative medicine from natural ingredients. The ethyl acetate fraction Phyllanthus niruri L. is suspected of having anticoagulant and thrombolytic activity due to the content of coumarin and flavonoid bioactive compounds. The purpose of this study was to determine the anticoagulant and thrombolytic activity of the ethyl acetate fraction of Phyllanthus niruri L. Anticoagulant activity test was carried out using the Lee-White method and the austek method by measuring CT (Clotting Time) and blood clotting. A thrombolytic activity test was performed by measuring the percentage of clot lysis. This study used a true experimental method with a Post Test Only Control Group Design model in 6 treatments, positive control, negative control and 4 samples, namely 250ppm, 500ppm, 1000ppm, 2000ppm. The results showed that the herb fraction Phyllanthus niruri L. had an anticoagulant activity with the largest CT value of 33.6 and the smallest area of blood clot 1.3% at a concentration of 2000ppm, while the results of the Kruskal Wallis test p<0.05, 0.005 which was a significant difference between treatments. In the thrombolytic activity test of ethyl acetate fraction of herb Phyllanthus niruri L., the largest clot lysis was 44.56% at 2000ppm compared to the positive control 46.33%, while the results of the ANOVA statistical test p<0.05, namely 0.017, which means there was a significant difference between treatments. Based on the results, it was known that the ethyl acetate fraction of the herb Phyllanthus niruri L. has anticoagulant and thrombolytic properties in vitro with the greater concentration was the greater effectiveness at the concentration measured in this study.
